Overview

CDK4 siRNA refers to a class of RNA interference-based therapeutics designed to silence the expression of cyclin-dependent kinase 4 (CDK4). CDK4 is a key regulator of the G1 to S phase transition in the cell cycle, and its overexpression is associated with various malignancies, including breast cancer, ovarian cancer, and non-small-cell lung carcinoma (NSCLC). By utilizing small interfering RNAs (siRNAs) delivered via various platforms such as lipid nanoparticles (LNPs), micellar nanoparticles (MNPs), or peptide-based systems, these agents induce the degradation of CDK4 mRNA, leading to cell cycle arrest and apoptosis in tumor cells. Research has also explored its use in synthetic lethal strategies, particularly in KRAS-mutant cancers, and in combination with other therapies like let-7 miRNA or CDK4 inhibitors like palbociclib.
