CDK6 Selective Oral SMi is an orally bioavailable small molecule inhibitor specifically targeting cyclin-dependent kinase 6 (CDK6), currently in preclinical development by Almac Discovery. CDK6 is a critical kinase that, in complex with D-type cyclins, phosphorylates the retinoblastoma (Rb) protein to facilitate the transition from the G1 to the S phase of the cell cycle. While many approved therapies are dual CDK4/6 inhibitors, this program focuses on achieving selectivity for CDK6, which may provide therapeutic benefits in specific cancer types or genomic backgrounds where CDK6 is the primary driver of proliferation. The program is being evaluated for a wide range of indications, including brain cancer, colorectal cancer, esophageal and gastric cancers, head and neck cancer, lung cancer, and various hematologic malignancies.
