CDKI-15 is a novel, highly selective small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). Developed by researchers at the University of South Australia, it demonstrates low nanomolar potency (Ki = 4 nM for CDK4) and high selectivity over other CDKs such as CDK1, CDK2, CDK7, and CDK9. In preclinical studies, CDKI-15 has shown the ability to reduce retinoblastoma (Rb) protein phosphorylation and induce G1 phase cell cycle arrest in cancer cells. It possesses excellent oral bioavailability (100% in mice) and has demonstrated significant anti-tumor activity in xenograft models of acute myeloid leukemia (AML), specifically the MV4-11 cell line, without detectable toxicity.
