CDN1163 is a small molecule allosteric activator of the sarco/endoplasmic reticulum Ca2+-ATPase 2 (SERCA2). It is designed to restore calcium homeostasis within the endoplasmic reticulum (ER), a process that is frequently disrupted in metabolic and neurodegenerative diseases. By enhancing SERCA2 activity, CDN1163 increases the transport of Ca2+ from the cytoplasm into the ER, thereby mitigating ER stress, improving mitochondrial function, and reducing hepatocellular inflammation. Preclinical research has demonstrated that CDN1163 can improve insulin sensitivity, reduce adiposity, and decrease the NAFLD activity score in mouse models of diet-induced obesity and steatohepatitis. While primarily studied in the context of metabolic disorders like nonalcoholic fatty liver disease (NAFLD) and type 2 diabetes, its mechanism of reducing ER stress has also led to investigation in neurodegenerative models such as Parkinson's disease.
