Overview

CDX-014 is an antibody-drug conjugate (ADC) designed for the treatment of cancer. It consists of a fully human immunoglobulin G1 monoclonal antibody targeting T-cell immunoglobulin mucin domain-containing protein 1 (TIM-1), a surface marker highly expressed in renal cell carcinoma and ovarian carcinoma. The antibody is covalently linked to the cytotoxic agent monomethyl auristatin E (MMAE) via a valine-citrulline linker. Upon binding to TIM-1 on tumor cells and internalization, MMAE is released inside the cell, leading to inhibition of tubulin polymerization and subsequent cytotoxic death of the cancer cell. CDX-014 was developed by Celldex Therapeutics using XenoMouse technology originally from Amgen/CuraGen Corporation. Clinical trials have shown manageable toxicity and early signs of antitumor activity in advanced refractory renal cell carcinoma[1][3][4][5].
