CDX-1140 + gemcitabine + nab-paclitaxel is an experimental combination regimen comprising a fully human CD40 agonist monoclonal antibody (CDX-1140), a nucleoside analog antimetabolite (gemcitabine), and a nanoparticle albumin-bound formulation of paclitaxel (nab-paclitaxel). CDX-1140 is designed to activate antigen-presenting cells such as dendritic cells and B cells by agonizing CD40, thereby enhancing antitumor immune responses independent of Fc receptor interactions. Gemcitabine inhibits DNA synthesis in rapidly dividing tumor cells, while nab-paclitaxel stabilizes microtubules, preventing cell division. This combination is being investigated for its potential synergistic immunotherapeutic and cytotoxic effects against advanced cancers. Early clinical studies are primarily in solid tumors, including pancreatic and biliary tract cancer, particularly for patients who have progressed on standard first-line therapies[1].
