Overview

CDX-1307 is a cancer vaccine and immunotherapy composed of a fully human monoclonal antibody (B11) targeting the mannose receptor on dendritic cells, genetically fused to the beta-subunit of human chorionic gonadotropin (hCG-beta), which serves as a tumor antigen. The drug is designed to selectively deliver hCG-beta to antigen-presenting cells, thereby stimulating robust humoral and cellular immune responses against hCG-beta-expressing epithelial cancers such as bladder, breast, colorectal, ovarian, and pancreatic cancers. The presence of hCG-beta in these tumors correlates with poor prognosis and may contribute directly to tumor progression. CDX-1307 has been evaluated in combination with immune adjuvants such as GM-CSF and Toll-like receptor agonists (poly-ICLC for TLR3; resiquimod for TLR7/8) to enhance its immunogenicity. Clinical studies demonstrated that CDX-1307 was well tolerated and induced significant immune responses but development was discontinued after early-phase trials[1][2][3][4][5][6].
