Overview

CDX-1307 + toll-like receptor agonists is an investigational combination immunotherapy consisting of CDX-1307, a fusion protein vaccine targeting the mannose receptor on antigen-presenting cells (APCs), fused to the human chorionic gonadotropin-beta subunit (hCG-β), and given together with toll-like receptor (TLR) agonists (specifically poly-ICLC, a TLR3 agonist, and resiquimod, a TLR7/8 agonist). CDX-1307 is designed to direct the tumor antigen hCG-β to dendritic cells through the mannose receptor, triggering both cellular and humoral immune responses against hCG-β-expressing cancers. The TLR agonists act as potent immunostimulatory adjuvants, enhancing vaccine-induced immune responses by activating innate immune pathways. This combination has been trialed primarily in patients with epithelial cancers such as bladder, breast, colorectal, pancreatic, and ovarian cancers[2][3][6][7].
