Overview

CDX-3379 (formerly KTN-3379) is a human immunoglobulin G1 lambda (IgG1λ) monoclonal antibody that selectively binds to a unique epitope on Receptor tyrosine-protein kinase erbB-3 (ErbB3, also known as HER3), locking the receptor in an inactive state and inhibiting both ligand-dependent and ligand-independent ErbB3 signaling. This inhibition prevents phosphorylation of ErbB3, thereby blocking downstream signaling pathways involved in cellular proliferation and survival of tumor cells expressing ErbB3. The drug was developed for the treatment of various cancers, including head and neck squamous cell carcinoma (HNSCC), thyroid cancer, malignant melanoma, and other solid tumors. It has been studied alone or in combination with cetuximab to overcome resistance mechanisms associated with EGFR-targeted therapies. CDX-3379 features half-life-extending YTE modifications in its Fc region for improved pharmacokinetics[1][2][4][5][6].
