Overview

CDX0239-PBD is a first-in-class antibody-drug conjugate (ADC) designed to target the anaplastic lymphoma kinase (ALK) receptor. It consists of a humanized anti-ALK monoclonal antibody (CDX0239) conjugated to a pyrrolobenzodiazepine (PBD) dimer, a potent cytotoxic payload. ALK is a receptor tyrosine kinase that is frequently overexpressed or mutated in various pediatric and adult malignancies, including neuroblastoma, fusion-positive rhabdomyosarcoma, and subsets of colorectal, breast, and ovarian carcinomas. Upon binding to ALK on the cell surface, the ADC is internalized, and the PBD dimer is released. The payload then binds to the DNA minor groove and forms covalent interstrand cross-links, which inhibit DNA replication and lead to cell death. CDX0239-PBD has demonstrated significant antitumor activity in preclinical models and is being developed for the treatment of ALK-positive cancers.
