CE-VST01-JC is an allogeneic, off-the-shelf T cell therapy developed for the treatment of progressive multifocal leukoencephalopathy (PML), a rare and often fatal demyelinating disease of the central nervous system caused by JC polyomavirus (JCPyV) in immunocompromised patients. The therapy consists of JCPyV-specific T cells expanded from healthy donors using a curated mixture of 36 JCPyV-derived peptides covering multiple viral antigens and HLA alleles, designed to maximize specificity and minimize graft-versus-host disease risk. These T cells display a polyfunctional profile with enrichment for stem-cell like memory phenotypes. CE-VST01-JC is being evaluated in ASCEND-JC, a global Phase 2 randomized, double-blind, placebo-controlled trial to assess its safety and efficacy in slowing or halting neurological progression in PML patients. There are currently no approved treatments for PML[1][3][4].
