Overview

CEA peptide-pulsed dendritic cells are an investigational cell-based immunotherapy in which autologous or allogeneic dendritic cells are loaded ex vivo with a synthetic cytotoxic T lymphocyte (CTL) epitope derived from the human carcinoembryonic antigen (CEA). The pulsed dendritic cells are then administered to patients to stimulate a tumor-specific immune response. The mechanism of action involves the presentation of the CEA-derived peptide by HLA class I molecules on the surface of mature dendritic cells, leading to activation and expansion of CD8+ cytotoxic T lymphocytes that recognize and kill CEA-expressing tumor cells. This approach is being studied primarily for cancers that overexpress CEA, such as colorectal cancer and other solid tumors. Clinical studies have demonstrated induction of specific anti-CEA immune responses in patients with resectable liver metastases from colorectal cancer[1][2][3][5].
