CEA peptide vaccines are investigational cancer immunotherapies designed to stimulate the immune system to recognize and attack tumor cells expressing carcinoembryonic antigen (CEA), a tumor-associated glycoprotein overexpressed in many cancers including colorectal, gastric, pancreatic, non-small cell lung, and breast carcinomas. These vaccines typically use synthetic peptides derived from immunodominant epitopes of the CEA protein. Some formulations involve pulsing patient-derived dendritic cells with CEA peptides to enhance T-cell activation and anti-tumor immunity[1][2][8]. The mechanism of action is based on presenting specific fragments of the CEA antigen to the immune system—often restricted by HLA haplotypes—to induce cytotoxic T lymphocyte responses against cancer cells expressing CEA[2][5]. Modified or agonist versions of these peptides have been developed to improve immunogenicity. Clinical trials have demonstrated that vaccination with these peptides is generally well tolerated and can elicit immune responses in patients with advanced cancers such as colorectal cancer[5].
