CEA-specific CAR-NK cells are an investigational chimeric antigen receptor (CAR) natural killer (NK) cell therapy targeting carcinoembryonic antigen (CEA), a glycoprotein overexpressed in colorectal cancer (CRC) and other solid tumors. The therapy involves engineering NK cells—either the NK-92 cell line or induced pluripotent stem cell (iPSC)-derived NK cells (iNKs)—to express a second-generation CAR, typically utilizing the high-affinity hMN-14 scFv. These cells may be further enhanced through cytokine engineering, such as the constitutive expression of interleukins, to boost their anti-tumor activity and persistence. Preclinical studies have demonstrated that these CAR-NK cells effectively reduce tumor progression in CEA-positive CRC xenograft models, presenting a potentially safer and more scalable alternative to CAR-T cell therapies.
