Overview

CEA-specific cytotoxic T lymphocytes (CTLs) are an autologous cellular immunotherapy developed by The First People's Hospital of Changzhou for the treatment of stage IV gastric cancer. The production process involves isolating a patient's peripheral blood mononuclear cells, differentiating them into dendritic cells (DCs), and transducing them with a recombinant adeno-associated virus (rAAV) carrying the carcinoembryonic antigen (CEA) gene. These CEA-expressing DCs are then used to prime and expand autologous T cells ex vivo, generating a population of CTLs specifically programmed to recognize and eliminate CEA-positive tumor cells. This approach leverages the high expression of CEA in gastric cancers to provide a targeted immune response against tumor cells while minimizing off-target effects.
