CEACAM5-MMAE protodrug is a preclinical antibody-drug conjugate (ADC) and protodrug being developed by Shasqi for the treatment of solid tumors. The therapeutic candidate targets Carcinoembryonic Antigen-Related Cell Adhesion Molecule 5 (CEACAM5), a cell-surface glycoprotein that is highly expressed in various epithelial cancers, including colorectal, pancreatic, and non-small cell lung cancer (NSCLC). The drug is built upon Shasqi's proprietary Click Activated Protodrugs Against Cancer (CAPAC) platform. Unlike traditional ADCs that rely on enzymatic cleavage or internalization for payload release, this "Click ADC" utilizes bioorthogonal click chemistry to trigger the release of the cytotoxic payload. The CEACAM5-MMAE protodrug consists of a CEACAM5-targeting antibody linked to a monomethyl auristatin E (MMAE) payload in an inactive protodrug form. The MMAE is selectively activated and released at the tumor site upon interaction with a chemical activator, thereby minimizing systemic exposure and potentially reducing the off-target toxicities typically associated with microtubule inhibitors.
