Cediranib + pemetrexed is an investigational combination therapy evaluated primarily in malignant pleural mesothelioma and non-small cell lung cancer. Cediranib is a potent oral small molecule inhibitor of vascular endothelial growth factor receptor (VEGFR), as well as platelet-derived growth factor receptor (PDGFR) and c-Kit, targeting tumor angiogenesis. Pemetrexed is an antifolate chemotherapy agent that inhibits multiple folate-dependent enzymes involved in purine and pyrimidine synthesis, disrupting tumor cell proliferation. The combination aims to enhance antitumor efficacy by combining cytotoxic and antiangiogenic mechanisms, and has been studied both as a doublet and as part of triplet regimens (with cisplatin), mainly for use in unresectable, chemotherapy-naïve malignancies such as malignant pleural mesothelioma[1][2][3][5].
