ceDNA vaccine is a non-viral genetic medicine platform developed by Generation Bio that utilizes closed-ended DNA (ceDNA) as its genetic payload. Unlike traditional plasmid DNA, ceDNA is a high-capacity, double-stranded, linear construct that is covalently closed, designed to provide stable gene expression without the use of viral vectors. The platform employs a proprietary vaccine-optimized cell-targeted lipid nanoparticle (ctLNP) delivery system to transport the ceDNA into specific cells, such as those in the liver or the immune system. An advanced iteration of this technology, known as immune-quiet DNA (iqDNA), is engineered to minimize detection by the host's innate immune system, thereby potentially increasing the durability and magnitude of the resulting humoral and T-cell responses. Generation Bio has partnered with Moderna to further develop this technology for various therapeutic and prophylactic applications.
