Overview

Cefalonium is a first-generation, semi-synthetic cephalosporin antibiotic belonging to the β-lactam class. It exhibits broad-spectrum bactericidal activity against Gram-positive and some Gram-negative bacteria, including *Staphylococcus aureus* (including penicillin-resistant strains), *Streptococcus* species, *Escherichia coli*, *Klebsiella* spp., and others commonly associated with bovine mastitis. Its primary mechanism of action is inhibition of bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), thereby interfering with the final phase of peptidoglycan synthesis and leading to cell lysis and death. Cefalonium is primarily used in veterinary medicine for routine dry cow therapy—to treat existing subclinical infections and prevent new intramammary infections during the dry period in dairy cattle.
