The protocol involves a combination of cefazolin, a first-generation cephalosporin administered intravenously at induction, and a subsequent one-week oral course of amoxicillin and clavulanic acid. Cefazolin and amoxicillin function by inhibiting bacterial cell wall synthesis through the targeting of penicillin-binding proteins (PBPs), while clavulanic acid serves as a beta-lactamase inhibitor to protect amoxicillin from enzymatic degradation by resistant bacteria. This specific sequential treatment strategy is being investigated in a Phase 4 clinical trial to compare its efficacy against a standard single-dose preoperative antibiotic prophylaxis.
