Cefazolin benzathine is a specific salt formulation of cefazolin, a first-generation cephalosporin antibiotic. It functions by binding to and inhibiting penicillin-binding proteins (PBPs), which are essential enzymes for bacterial cell wall synthesis, thereby leading to bacterial cell lysis. While the sodium salt of cefazolin is the standard form used in human clinical practice for surgical prophylaxis, the benzathine salt is more commonly associated with veterinary applications or long-acting formulations. The drug was studied in a Phase 4 pharmacokinetic trial led by Hôpital Erasme to determine optimal dosing strategies in obese versus non-obese adult surgical patients. This research aimed to ensure that prophylactic concentrations remain above the minimum inhibitory concentration (MIC) for target pathogens throughout the duration of surgery, addressing the physiological changes in drug distribution and clearance associated with obesity.
