Overview

Cefotiam hexetil is an orally active, second-generation cephalosporin antibiotic and a prodrug of cefotiam. It was originally developed by Takeda and is characterized by its broad-spectrum activity against both Gram-positive and Gram-negative bacteria, including *Staphylococcus aureus*, *Streptococcus pneumoniae*, *Escherichia coli*, and *Haemophilus influenzae*. Upon oral administration, cefotiam hexetil is rapidly absorbed and hydrolyzed by esterases in the intestinal wall and blood to release the active moiety, cefotiam. Its mechanism of action involves the inhibition of bacterial cell wall synthesis through binding to penicillin-binding proteins (PBPs), which leads to cell lysis and death. It is primarily indicated for the treatment of respiratory tract infections, urinary tract infections, and skin and soft tissue infections.
