VNRX-7145 is an orally bioavailable, broad-spectrum boronic acid beta-lactamase inhibitor (BLI) developed by Venatorx Pharmaceuticals. It serves as a prodrug of the active inhibitor VNRX-5232. VNRX-7145 is designed to be administered in combination with a partner beta-lactam antibiotic, specifically ceftibuten, to address the clinical challenge of multi-drug resistant (MDR) Gram-negative bacteria. The compound targets a wide range of beta-lactamases, including Class A (ESBLs, KPC), Class C (AmpC), and Class D (OXA-48) enzymes. By inhibiting these enzymes, VNRX-7145 restores the antibacterial activity of ceftibuten against resistant strains, including Carbapenem-Resistant Enterobacteriaceae (CRE). The combination is primarily being developed as an oral treatment option for complicated urinary tract infections (cUTI) and other serious infections that currently necessitate intravenous therapy.
