CEL-5000 is a peptide-based immunomodulator developed by CEL-SCI Corporation for the treatment of rheumatoid arthritis. It utilizes the proprietary Ligand Epitope Antigen Presentation System (LEAPS) technology, which involves the construction of a chimeric peptide containing two distinct functional domains: a T-cell binding ligand (derived from the human MHC class II molecule) and a disease-specific antigen epitope (derived from type II collagen). The mechanism of action involves the targeted delivery of the collagen epitope to T-cells in a manner that promotes the induction of a protective, anti-inflammatory immune response, potentially by shifting the T-cell cytokine profile from Th1 to Th2 or by inducing regulatory T-cells. Although CEL-5000 demonstrated efficacy in preclinical models of collagen-induced arthritis, the program was later superseded by CEL-1000, a refined LEAPS candidate for the same indication.
