CEL_Amide

CEL_Amide is a **novel small molecule inhibitor** specifically targeting **LIM domain kinase 1 and 2 (LIMK1/2)**. It has shown **preclinical anti-leukemic activity** in experimental models of BCR::ABL+ acute lymphoblastic leukemia (ALL). Mechanistically, CEL_Amide's inhibition of LIMK1/2 is thought to disrupt the ROCK-LIMK-cofilin signaling axis, which plays a critical role in actin cytoskeleton dynamics and, indirectly, Aurora kinase A activation. This action can sensitize leukemic cells to apoptosis. CEL_Amide demonstrates strong **synergy with BCR::ABL-targeting tyrosine kinase inhibitors (TKIs)** in vitro and in vivo, suggesting it could help overcome resistance to existing TKIs, including in cells harboring TKI resistance mutations.
