This is a combination regimen consisting of three drugs—celecoxib, cisplatin, and irinotecan—used primarily in investigational settings for the treatment of certain cancers. - **Celecoxib** is a selective cyclooxygenase-2 (COX-2) inhibitor that reduces prostaglandin synthesis and has anti-inflammatory as well as potential antitumor effects. - **Cisplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. - **Irinotecan** is a topoisomerase I inhibitor that prevents DNA unwinding and replication in cancer cells. The rationale for combining these agents includes the radiosensitizing effect of irinotecan and its synergy with cisplatin; celecoxib may enhance antitumor efficacy by inhibiting COX-2-mediated tumor growth and reducing some toxicities associated with chemotherapy or chemoradiation[1][4][6]. This combination has been studied mainly in phase I trials for locally advanced non-small cell lung cancer (NSCLC) and esophageal cancer[1][2][3][4]. The addition of celecoxib aims to improve outcomes without increasing normal tissue toxicity.
