Overview

This is a combination regimen consisting of four drugs: - Celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor with anti-inflammatory and potential antineoplastic properties. - Fluorouracil (5-FU), an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis in rapidly dividing cells. - Leucovorin (folinic acid), which enhances the binding of 5-FU to thymidylate synthase, increasing its cytotoxicity. - Oxaliplatin, a platinum-based chemotherapeutic agent that forms DNA crosslinks and inhibits DNA replication and transcription. This combination has been studied primarily in colorectal cancer. The addition of celecoxib to standard FOLFOX chemotherapy regimens has been evaluated for its potential to reduce recurrence risk or improve outcomes in stage III colon cancer and metastatic colorectal cancer. Clinical trials have shown that while the safety profile is acceptable, there may be limited synergy between celecoxib and FOLFOX[1][2][6].
