Cemacabtagene ansegedleucel is a next-generation, allogeneic chimeric antigen receptor T cell (CAR-T) therapy targeting CD19, developed for the treatment of large B-cell lymphoma (LBCL), including diffuse large B-cell lymphoma, primary mediastinal B-cell lymphoma, marginal zone B-cell lymphoma, follicular lymphoma, and relapsed/refractory chronic lymphocytic leukemia (CLL). Unlike autologous CAR-T therapies that use a patient’s own cells, this “off-the-shelf” product uses donor-derived T cells engineered to express an anti-CD19 CAR. The therapy incorporates gene edits to knock out the T cell receptor gene—reducing risk of graft-versus-host disease—and the CD52 gene—conferring resistance to anti-CD52 antibody treatments used for host immune suppression. Cemacabtagene ansegedleucel has demonstrated durable responses in clinical trials and is currently in Phase II development for LBCL and CLL[1][2][3][4][5][8].
