Overview

A combination regimen consisting of **cemiplimab**, an immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1); **carboplatin**, a platinum-based chemotherapeutic agent; and **paclitaxel**, a taxane-based microtubule inhibitor. Cemiplimab enhances anti-tumor immunity by blocking PD-1, allowing T-cell activation against cancer cells[7][1][5]. Carboplatin forms DNA adducts, causing apoptosis in rapidly dividing tumor cells; paclitaxel stabilizes microtubules to inhibit mitosis, inducing cell death[1][4][7]. This regimen has been studied in locally advanced or metastatic non-small cell lung cancer (NSCLC)[7][5][3], recurrent/metastatic squamous cell carcinoma of the head and neck[1], and other solid tumors. The combined approach offers improved efficacy compared to chemotherapy alone, at the cost of increased immune and hematologic toxicity[7][5][3][4].
