Cenerimod is an orally available, potent, and highly selective small molecule modulator of the sphingosine-1-phosphate receptor 1 (S1P1). It is under clinical development primarily for the treatment of systemic lupus erythematosus (SLE), a chronic autoimmune disease. Cenerimod works by selectively binding to S1P1 receptors on lymphocytes, sequestering them in lymph nodes and thereby reducing their circulation in the bloodstream. This mechanism reduces immune-mediated tissue damage characteristic of SLE. Unlike other S1P receptor modulators, cenerimod displays unique biased signaling properties at S1P1 that may contribute to a favorable safety profile with reduced cardiovascular and pulmonary side effects. The drug is being developed by Idorsia and has reached Phase 3 clinical trials for moderate to severe SLE[3][5][6][8].
