Overview

A combination therapy using **CEP-9722**, an oral pro-drug of the PARP-1 and PARP-2 inhibitor CEP-8983, with **temozolomide**, an alkylating agent. CEP-9722 inhibits poly(ADP-ribose) polymerase 1 and 2, impairing DNA repair and thereby sensitizing tumor cells to DNA-damaging chemotherapy. Temozolomide induces apoptosis by methylating DNA. The combination is investigated for its ability to enhance antitumor activity and overcome resistance to temozolomide in advanced solid tumors. Phase I studies established the maximum tolerated dose (MTD) and characterized pharmacokinetics, safety, and preliminary efficacy for this regimen[1][3][5].
