Overview

**CEQ508** is an investigational orally administered RNAi therapeutic utilizing Cequent's transkingdom RNAi (tkRNAi) platform. It consists of engineered attenuated bacteria that deliver short-hairpin RNA (shRNA) specifically to dysplastic intestinal tissue, silencing **beta-catenin** (CTNNB1) mRNA to reduce protein levels in epithelial cells. Developed initially by Cequent Pharmaceuticals and Marina Biotech for **Familial Adenomatous Polyposis (FAP)**, a condition with dysregulated beta-catenin leading to colonic polyps and colorectal cancer risk, it received FDA IND approval, Fast Track designation, and advanced to Phase 2 trials, though development appears stalled.[1][2][4][7][16]
