CER-1236 is a novel chimeric antigen receptor (CAR) T cell therapy developed by CERo Therapeutics. It is the first CAR-T therapy designed to target Tim-4L and uniquely incorporates programmed phagocytic activity into T cells, enabling both direct cytotoxicity and phagocytic clearance of tumor cells. The extracellular domain of TIM-4, which recognizes the "eat me" signal phosphatidylserine, is fused with intracellular signaling domains (TLR2/TIR, CD28, and CD3ζ), enhancing both phagocytosis and T cell-mediated killing. Preclinical studies have demonstrated that CER-1236 can selectively kill ovarian cancer cells without toxicity to healthy tissues and shows promise in models of non-small cell lung cancer (NSCLC) and mantle cell lymphoma (MCL). The dual mechanism may help overcome resistance barriers seen in solid tumor CAR-T trials. Phase 1 clinical trials are underway for advanced solid tumors including NSCLC and ovarian cancer as well as for hematological malignancies such as acute myeloid leukemia (AML)[1][2][3][4][5][6][8].
