Ceramide analog 315, chemically known as (S,E)-3-hydroxy-2-(2-hydroxybenzylidene)amino-N-tetradecylpropanamide, is a novel synthetic ceramide-mimicking small molecule developed by researchers at the Xavier University of Louisiana. It is designed to target the ceramide signaling pathway for the treatment of various cancers, particularly chemo-resistant breast cancer and primary effusion lymphoma (PEL). Ceramide analog 315 acts as an acid ceramidase inhibitor, preventing the breakdown of tumor-suppressive ceramides into pro-survival sphingosine-1-phosphate (S1P). By increasing intracellular ceramide levels, it induces mitochondrial toxicity, cytochrome c release, caspase-9 activation, and subsequent apoptosis. In preclinical models, ceramide analog 315 has demonstrated significant anti-proliferative, pro-apoptotic, and anti-tumor activities, reducing tumor burden and preventing metastasis in vivo.
