Ceramide nanoliposome is a lipid-based nanoparticle formulation in which the bioactive sphingolipid ceramide is encapsulated within nanoliposomes. This design overcomes the natural cell impermeability and poor solubility of ceramide, enabling its delivery to tumor cells. The drug induces apoptosis and non-apoptotic cytotoxicity in cancer cells by modulating key signaling pathways, including inhibition of glycolysis (via Glyceraldehyde 3-phosphate dehydrogenase downregulation) and overcoming drug resistance mechanisms such as Myeloid cell leukemia sequence 1 protein expression. It has demonstrated antineoplastic activity in preclinical models for various cancers, including liver cancer, pancreatic cancer, leukemia (including acute myeloid leukemia and chronic lymphocytic leukemia), breast cancer, and solid tumors. The formulation was originally developed by Keystone Nano (now PendreaBio) with academic collaboration from the University of Virginia[1][3][4][5][6].
