Cergutuzumab amunaleukin is a recombinant fusion protein designed for cancer immunotherapy. It consists of a genetically engineered human IgG1 kappa monoclonal antibody (cergutuzumab) targeting carcinoembryonic antigen related cell adhesion molecule 5 (CEA/CEACAM5/CD66e), fused to an engineered interleukin-2 variant (IL-2v) with abolished CD25 binding. This design aims to selectively activate immune effector cells over regulatory T-cells and enhance tumor targeting by binding to CEA-expressing cancer cells while avoiding Fc-mediated effector functions and reducing off-target effects. The drug is being developed primarily for the treatment of advanced or metastatic solid tumors that express carcinoembryonic antigen related cell adhesion molecule 5 (CEA). Its mechanism involves both antagonism of carcinoembryonic antigen related cell adhesion molecule 5 (CEACAM5) and agonism at the interleukin-2 receptor beta/gamma subunits, stimulating anti-tumor immune responses. Developed by Roche/Genentech, it has been evaluated in Phase 1 clinical trials as monotherapy and in combination with atezolizumab for patients whose disease progressed on standard therapies.
