CERIG siRNA is a small interfering RNA (siRNA) designed to silence the expression of Cancer ER Invasion Gene (CERIG), also known as KIAA1199 or CEMIP (Cell Migration-Inducing Protein). CERIG is a novel endoplasmic reticulum (ER) resident protein that is upregulated in human invasive breast cancer and other solid tumors, where its expression correlates with poor prognosis. Mechanistically, CERIG forms a complex with the ER chaperone GRP78/BiP and promotes ER calcium leakage, leading to increased cytoplasmic calcium levels and subsequent activation of the PKC-MAPK pathway, which drives cancer cell migration, invasion, and metastasis. Silencing of CERIG using CERIG siRNA in aggressive cancer cells disrupts mesenchymal morphogenesis, induces a mesenchymal-to-epithelial transition (MET), and inhibits cancer cell migration and metastasis in vitro and in vivo.
