ceritinib + docetaxel

This combination therapy consists of ceritinib and docetaxel, currently being evaluated in a Phase I/Ib clinical trial (NCT03611738) led by the H. Lee Moffitt Cancer Center and Research Institute. Ceritinib is an oral small molecule tyrosine kinase inhibitor that primarily targets anaplastic lymphoma kinase (ALK), but also inhibits ROS1 and the insulin-like growth factor 1 receptor (IGF-1R). Docetaxel is a taxane-based chemotherapy agent that works by binding to and stabilizing tubulin, which disrupts microtubule assembly and leads to cell cycle arrest and apoptosis. The study investigates the potential of ceritinib to enhance the efficacy of docetaxel in patients with advanced non-small cell lung cancer (NSCLC) who are ALK-negative and EGFR wild-type, a population that has progressed on platinum-based chemotherapy and for whom docetaxel is a standard but often insufficiently effective second-line treatment.
