Drug intelligence / Profile preview
Cesalatamig (AI-081) is an investigational, potentially best-in-class bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF). Developed by OncoC4, the biologic is designed with a head-and-tail bispecific configuration using proprietary high-affinity anti-PD-1 (AI-025) and anti-VEGF (AI-011) antibodies. It features Fc-silencing modifications to prevent the depletion of PD-1-positive effector T cells and exhibits strong cooperative binding in tumor microenvironments enriched with both targets. Cesalatamig is currently being evaluated in Phase 1/2 clinical trials (such as the BIPAVE-001 study) for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC) in patients whose disease has progressed following PD-(L)1-targeted immunotherapy and platinum-based chemotherapy. The U.S. FDA has granted Fast Track Designation to cesalatamig for this indication.
