Cethrin is a recombinant protein therapeutic derived from the bacterial enzyme C3 transferase. It acts as an inhibitor of Rho proteins by covalently modifying and inactivating them, thereby blocking Rho-mediated axonal growth cone collapse and inhibiting neuronal apoptosis after spinal cord injury (SCI). By inhibiting Rho kinase activity at the site of injury, Cethrin promotes regeneration of cut axons and remodeling of damaged neural circuits. The drug is delivered topically during decompression surgery for acute traumatic cervical spinal cord injury. Its primary indication is for acute traumatic cervical SCI in adolescents and adults. Clinical trials have reached phase II/III development status but it does not currently have marketing authorization in any country[1][3][5][8].
