CETi-1 is a therapeutic vaccine candidate designed to treat hypercholesterolemia and atherosclerosis by inducing the production of endogenous antibodies against cholesteryl ester transfer protein (CETP). CETP is a plasma glycoprotein that mediates the transfer of cholesteryl esters from high-density lipoprotein (HDL) to pro-atherogenic lipoproteins such as VLDL and LDL. By inhibiting CETP activity, CETi-1 aims to raise HDL cholesterol levels and improve the lipid profile to reduce cardiovascular risk. The vaccine was developed by Avant Immunotherapeutics (now Celldex Therapeutics) and consists of a synthetic peptide corresponding to the C-terminal catalytic site of CETP, conjugated to a carrier to overcome self-tolerance. Clinical development reached Phase 2, but the program was ultimately discontinued.
