Overview

Cetirizine is a second-generation antihistamine used to relieve symptoms of allergic rhinitis and urticaria by selectively antagonizing the histamine H1 receptor, thereby reducing the effects of endogenous histamine. It is widely marketed under the brand name Zyrtec and others for allergy relief[3]. JNJ-39758979 is a selective, high-affinity small molecule antagonist of the histamine H4 receptor, developed by Janssen (Johnson & Johnson). It was investigated for its anti-inflammatory and antipruritic properties in conditions such as pruritus, asthma, atopic dermatitis, and rheumatoid arthritis. Clinical development was halted during phase II due to cases of idiosyncratic drug-induced agranulocytosis[1][2][6][7]. The combination "cetirizine + JNJ-39758979" refers to co-administration studied in clinical trials for their potential additive or synergistic effects on inhibiting histamine-induced pruritus; both act via different histamine receptors (H1 vs H4)[2].
