Cetuximab + capecitabine is a combination regimen used primarily in the treatment of metastatic colorectal cancer, particularly in patients with RAS or BRAF wild-type tumors. Cetuximab is a chimeric monoclonal antibody that targets the extracellular domain of the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in cell proliferation, survival, and angiogenesis. Capecitabine is an oral prodrug that is enzymatically converted to 5-fluorouracil (5-FU) in tumor tissue; 5-FU inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. This combination has been studied as both first-line therapy and as maintenance therapy after induction chemotherapy for metastatic colorectal cancer. Clinical trials have shown that this regimen can achieve meaningful progression-free survival with manageable toxicity profiles[1][3][6].
