Overview

Cetuximab + carboplatin + gemcitabine is a combination regimen used primarily in the treatment of advanced non-small cell lung cancer (NSCLC) and has also been studied in other solid tumors. Cetuximab is a chimeric monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in tumor cell proliferation and survival. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to apoptosis. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, resulting in cytotoxicity particularly during the S phase of the cell cycle. The combination aims to enhance antitumor efficacy through complementary mechanisms: cetuximab blocks EGFR-mediated growth signals, while carboplatin and gemcitabine induce direct cytotoxic effects on tumor cells[1][2][3]. This regimen has demonstrated acceptable toxicity profiles and encouraging response rates in clinical studies for chemotherapy-naïve patients with EGFR-positive stage IV NSCLC[1].
