Cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), is combined with platinum-based chemotherapy agents (cisplatin or carboplatin) and fluorouracil for the first-line treatment of recurrent or metastatic squamous cell carcinoma of the head and neck. Cetuximab inhibits EGFR-mediated signaling, leading to reduced tumor cell proliferation and survival. Cisplatin and carboplatin are platinum-containing chemotherapeutic agents that cause DNA crosslinking and apoptosis, while fluorouracil is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis. This combination has been shown to improve overall survival compared to chemotherapy alone in this patient population[1][2][3].
