Cetuximab + dalotuzumab + irinotecan is an investigational combination therapy studied primarily in metastatic colorectal cancer, especially in patients with wild-type KRAS tumors. Cetuximab is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling to inhibit tumor cell proliferation. Dalotuzumab (MK-0646) is a humanized monoclonal antibody that targets the insulin-like growth factor 1 receptor (IGF-1R), inhibiting IGF-1-mediated signaling involved in tumor growth and survival. Irinotecan is a small molecule chemotherapeutic agent that inhibits DNA topoisomerase I, leading to DNA damage and apoptosis of rapidly dividing cells. The triple regimen was evaluated for safety and efficacy but did not show improved survival compared to standard regimens; it was generally well tolerated[1][2][3][4].
