Overview
Cetuximab + platinum + 5-fluorouracil is a **combination regimen** comprised of three agents: **cetuximab** (a chimeric monoclonal antibody targeting EGFR), a **platinum-based chemotherapy** (commonly cisplatin or carboplatin), and **5-fluorouracil** (a pyrimidine analog antimetabolite). Cetuximab inhibits the epidermal growth factor receptor (EGFR), blocking downstream signaling that drives cancer cell proliferation and survival. Platinum agents form DNA crosslinks and disrupt DNA synthesis/repair, leading to cancer cell death. 5-fluorouracil inhibits thymidylate synthase and disrupts DNA/RNA synthesis. This regimen was approved by the FDA as first-line treatment for patients with recurrent locoregional or metastatic squamous cell carcinoma of the head and neck (SCCHN), and it prolongs survival compared to chemotherapy alone[5][4].
