Cetuximab + RT is a combination therapeutic regimen consisting of the EGFR-targeting monoclonal antibody cetuximab and ionizing radiation (radiotherapy). Cetuximab is a chimeric IgG1 monoclonal antibody that binds to the extracellular domain of the human epidermal growth factor receptor (EGFR), which is frequently overexpressed in squamous cell carcinoma of the head and neck (SCCHN). By blocking ligand binding, cetuximab inhibits downstream signaling pathways involved in cell proliferation and survival, induces receptor internalization, and promotes antibody-dependent cell-mediated cytotoxicity (ADCC). When combined with radiotherapy, cetuximab acts as a radiosensitizer, enhancing the anti-tumor effects of radiation. This regimen is specifically indicated for the treatment of locally advanced SCCHN.
