Cetuximab + taxane + carboplatin is a combination chemotherapy regimen composed of cetuximab (a monoclonal antibody targeting EGFR), a taxane (typically paclitaxel or docetaxel, both microtubule-stabilizing small molecules), and carboplatin (a platinum-based alkylating agent). Cetuximab binds to the extracellular domain of the epidermal growth factor receptor (EGFR), inhibiting downstream signaling and cell proliferation. Taxanes disrupt microtubule dynamics, leading to mitotic arrest and apoptosis. Carboplatin forms DNA crosslinks, inhibiting DNA synthesis and function. This regimen has been studied as induction or first-line chemotherapy primarily for squamous cell carcinoma of the head and neck, non-small-cell lung cancer, and carcinoma of unknown primary. Dosing regimens vary, with weekly and every-3-week schedules documented. The combination is considered for its efficacy and potentially lower toxicity compared to certain other regimens, but randomized studies show mixed results in survival benefits over chemotherapy alone[1][2][3][4][5].
