Cetuximab-conjugated gold nanospheres are an experimental nanomedicine construct in which spherical gold nanoparticles are covalently functionalized on their surface with the anti-EGFR monoclonal antibody cetuximab (also known as C225) to enhance targeted delivery and antitumor efficacy. By combining the high-affinity EGFR binding and signaling inhibition of cetuximab with the multivalent display, cellular uptake, and optional radiosensitizing or photothermal properties of gold nanospheres, these conjugates increase EGFR internalization, promote receptor degradation, and more potently suppress downstream signaling pathways such as RAS/RAF/MEK/ERK and PI3K/AKT compared with cetuximab alone in EGFR-positive tumors including non-small cell lung, colorectal, pancreatic, and head and neck cancers. Preclinical studies demonstrate enhanced cytotoxicity, reduced tumor growth, and, when combined with radiotherapy or other external energy sources, increased tumor radiosensitization and hyperthermic cell killing, while leveraging the relative biocompatibility and tunable size/surface chemistry of gold nanoparticles as a delivery platform.
